PhD project description
Project Objectives:
- Evaluate amount and quality of human circulating RNAs and assess possible applications of epitranscriptomics profiling.
- Optimise all steps of RNA extraction, chemical treatment and library preparation steps for reliable deep sequencing of target RNAs.
- Establish technology allowing robust and precise epitranscriptomic profiling of RNA species present in human liquid biopsies and body fluids like blood, saliva and urine samples.
- Apply the developed technology to profiling of the cohort of PCa cancer patients and compare them to healthy subjects.
- Explore possibility for coupling this analysis to nanopore sequencing technology.
Your role:
- Gain the knowledge on the composition of stable non-coding RNAs in human liquid biopsies and body fluids and select suitable and relevant targets for epitranscriptomic profiling.
- Establish reference RNA modification profiles for healthy subjects.
- Compare RNA modification profiles for healthy subjects and PCa patients.
- Evaluate the suitability of RNA epitranscriptomic profiles as ONT biomarkers of PCa.
