PhD project description
The proposed PhD project aims to:
- Investigate the effect of Nm perturbation at selected specific ribosomal sites on cancer cell invasive behaviour by generating stable knockout and overexpression 2D, 3D cell lines and xenograft models.
- Analyse the downstream molecular pathways by which specific Nm marks exercise their function to unveil their role in ribosome function and PCa progression.
- Develop vectors targeting snoRNAs/Nm tandems relevant in PCa progression.
Expected results:
Perturbation of ribosomal 2’O-methylation (Nm) leads to deficient function of cancer ribosomes (translation) and our recent data supports the functional involvement of individual Nm marks in PCa tumour growth.
- Generated molecular and cellular tools to study the effects of Nm perturbation on cancer cells.
- Phenotypic characterization of the obtained cell models
- Described the role of selected Nm marks in ribosomal function and their relevance in PCa progression.
